Walnut Husk Ethanol Extract Possess Antioxidant Activity and Inhibitory Effect of Matrix Metalloproteinase-1 Expression Induced by Tumor Necrosis Factor Alpha in Human Keratinocyte

Abstract
This study was evaluated antioxidant activity of ethanol extracts of Juglans regia (walnut) husk (WHE) using 1,1-diphenyl-2-picryl hydrazyl (DPPH) method. Also, cell viability of WHE on human keratinocyte (HaCaT) was evaluated by sulforhodamine B (SRB) assay. Inhibitory effect of WHE on tumor necrosis factor alpha (TNF-¥á) induced Matrix Metalloproteinase-1 (MMP-1 ) expression in HaCaT was analyzed by reverse transcriptase polymerase chain reaction (RT-PCR). As the results, WHE was showed similar antioxidant activity to vitamin C at concentration of 6.25, 12.5, 25, 50 ¥ìg/mL. WHE of low concentration (6.25, 12.5 and 25 ¥ìg/mL) didn¡¯t exhibited cytotoxicity in HaCaT. However, treatment of 50 and 100 ¥ìg/mL WHE were showed cell viability of about 98.4% and 91.1%, respectively, in same cells. Matrix Metalloproteinase-1 (MMP-1 ) mRNA were induced by TNF-¥á in HaCaT cells and WHE treatment of 10, 20, 50 and 100 ¥ìg/mL were decreased MMP-1 mRNA with dose-dependent manner. Exhibiting excellent antioxidant activity with high cell viability, and potent inhibitory effect on MMP-1 mRNA, ethanol extracts of walnut husk extracts can be applied in cosmetics as a MMP -1 inhibitor.
